The Leica EM AFS2 is a specially designed tool dedicated to low temperature sample fixation and resin embedding. By doing specimen preparation at low temperatures, artifacts such as sample shrinkage, membrane movement and sample extraction are greatly reduced when compared to specimens processed by standard room temperature electron microscopy protocols.
The Leica AFS2 is equipped with a stereomicroscope, fume exhaust system for added safety, fast LED UV polymerization for Lowicryl resin polymerization, and an intuitive programmable interface. For processing protocols that do not require the use of Osmium Tetroxide, an automated robotic system is available to do all solution changes.
Freeze substitution protocols for bacteria, plant samples, tissue culture cells, cultured cells grown on aclar dics and samples for correlative light and electron microscopy (CLEM) as well as immunogold labeling have been tested and implemented by the electron microscopy facility.
