The Tokuyasu method is an electron microscopy technique that has been developed for improved Immunolabeling.This technique involves infiltrating a sample with sucrose. The sucrose, which is a cryo protectant, will prevent ice crystal formation when the sample is frozen. The sample is then cut in a frozen state before being warmed up to room temperature prior to the immunolabelling protocol. The majority of sample alterations that occur in the convention electron microscopy protocol are prevented because of the absence of heavy sample fixation and dehydration, allowing a better recognition between antigen and antibody.
