(*) The 63x objective has a correction ring for different imersion media. It can be used with water or glycerol, but it has to be corrected for which to use. Speak with the UIC personnel if you need to change it.

Setup

Turn on procedures

  1. Turn on Definite Focus control box (if this is not the first, there might be problems with the this function).
  2. Turn on Colibri control box (LED fluorescent light source).

    • You have to turn on the LEDs you are going to use and set their intensity.
  3. Turn on the microscope control box.
  4. Turn on the microscope (button on the left side of the microscope body).
  5. Turn on the XY-stage control box.
  6. Turn on the camera.
  7. Start the MicroManager software.
  8. Set up your experiments on the microscope.

Turn off procedures

  1. Turn off the microscope control box.
  2. Turn off the camera.
  3. Turn off the Colibri control box.
  4. Turn off the XY-stage control box.
  5. Turn off the Definite Focus control box.
  6. If you used any immersion lenses be sure to clean them.
  7. TAKE OUT EVERYTHING YOU BROUGHT!, eg, pipettes, slides, etc...

Acquisition

  • Grid Colection / Tile

    It is possible to acquire grids and tiles of big samples through Zen software. For a more detailed information, please check the following link or click in the image to the right.

  • Tile Scan

  • Definite Focus

    Definite Focus is Zeiss's version of Autofocus through hardware. This funcionality allows continuous focus of the sample through coverslip position, keeping the sample focused during the whole acquisition with the main objective of preventing drift in Z over long periods of time.
